利用染料結(jié)合的方法來(lái)定量檢測(cè)釋放到組織培養(yǎng)液或從生物材料中提取的彈性蛋白。
 
應(yīng)用范圍:樣本可來(lái)源于細(xì)胞培養(yǎng)基和含細(xì)胞外基質(zhì)成分的組織提取物(如彈性軟骨、主動(dòng)脈、皮膚等),可檢測(cè)可溶性彈性蛋白原和不可溶性彈性蛋白(抽提后以可溶性彈性蛋白多肽的形式檢測(cè))。
 
樣本要求:樣本體積5-100ul,包含5-75ug的彈性蛋白
 
實(shí)驗(yàn)操作步驟

彈性蛋白的分離:用彈性蛋白沉淀劑處理樣本,使彈性蛋白沉淀下來(lái)。
彈性蛋白與染料結(jié)合:裝有沉淀的彈性蛋白管中,加染料和反應(yīng)液,反應(yīng)60min以上
彈性蛋白-染料復(fù)合物獲得:去掉未結(jié)合的染料
復(fù)合物染料的釋放:添加染料釋放劑,使彈性蛋白-染料復(fù)合物中的染料釋放出來(lái)
釋放出的染料檢測(cè):繪制標(biāo)準(zhǔn)曲線(xiàn),513nm或405nm處定量檢測(cè)
